ABSTRACT

Hirudo nipponica Whitman,a Chinese medicine which has a strong effect on platelet aggregation and antithrombin activity,is involved in the studies. The current situation is adverse to the cultivation, development and utilization of H.nipponica.Prior to summarizing the current research status and the unresolved problems and the prospective of H.nipponica, domestic and foreign literatures over the H.nipponica was discussed and judged, aiming at providing a reference for the comprehensive development and utilization of H.nipponica.

ABSTRACT

To address the role of Pr1 gene in the process of Hirsutella sinensis infecting Hepialus gonggaensis, differential expression of subtilisin-like protease gene was detected. In the present study, Pr1 gene analogues from H. sinensis were obtained by PCR strategy using specific primers designed from conserved regions of Pr1 gene reported in the GenBank. Then we detected the changes in the expression of Pr1 gene before and after infecting H. gonggaensis using real-time quantitative PCR. We obtained the partial sequence of Pr1 gene with the length of 535 bp (GenBank accession: KC009680). Real-time PCR results showed that the expression level of Pr1 gene was significantly different among 8 samples (P < 0.01). Pr1 gene showed the obvious higher expression level (2-3 folds) after infecting the H. gonggaensis, suggesting that the Pr1 gene may play an important role in the process of H. sinensis infecting H. gonggaensis. The present study paves a way for further identification on infectivity assessment of H. sinensis.

ABSTRACT

OBJECTIVETo clarify the relevance of amino acid content in cultivated Cordyceps and cultivation materials.

METHODThe content of amino acid was determined with L-8800 amino acid analyzer, and the relevance of amino acid content was analyzed with SPSS.

RESULT AND CONCLUSIONExcept mycelium of the C. sinensis or the blood-lymph of the larva, the cultivated Cordyceps and the main relevant cultivation materials had detected to contain all kinds of amino acids. Except among the mycelium, the blood-lymph of the larva, the part of the larva or of the stroma of cultivated Cordyceps, there was distinct relevance of amino acid contents in cultivated Cordyceps and the cultivation materials (P<0.01).

ABSTRACT

OBJECTIVETo study the biological character of Hepialus introduced from Yunnan province.

METHODH. jianchuanensis introduced from Yunnan was bred artificially to observe its biological character.

RESULTThe pupa duration was about 40 d, pupa emergence lasted for one day, female to male ratio was about 1.2:1, the mating peak was 19:30-20:30, the adults duration was about 25 d, the average life time of female was 5-6 d and that of male 6-7 d, the average number of the producing egg was about 300, the average weight of the 100 eggs was 10.33 mg. Lasting period of the larva is 350 d and the infancy larva showed the character of the polyphagy and cold endurance.

CONCLUSIONResults indicate that there are some differences in the whole duration, weight of the 100 eggs and the number of the eggs left in the abdomen between H. jianchuanensis and others bred at the Kangding base.

ABSTRACT

The particularities of oil suspension formulation can raise the invasive rate of Hirsutella sinensis to host of Hepialidae for the commercialization of artificial cultivation of Cordyceps sinensis. So it is important to develop a high cell viability oil suspension formulation of C. sinensis spawn. According to the characteristics of the oil suspension formulation, MTT assay is adapted and optimized. The result is as follows: reaction time 120 min, reaction temperature 37?C, methylbenzene as extracting agent, and a positive linear correlation established between active cell weights and cell viability. Varieties and concentrations of assistance agents in oil suspension formulation have been selected with the refined MTT assay, and further optimized together with cell concentrations through orthogonal experiment. The optimal combination project was obtained, namely, cell concentration 0.15 g/mL, aluminium stearate 60 mg/mL, and SPAN-80 50 ?L/mL. Results of stability test on the oil suspension formulation indicate that cell viability can maintain above 90% at 4?C after one month.

ABSTRACT

OBJECTIVE:To gain clear idea of influence of multiorigins of Cordyc ep s on quality and nature of this drug.METHODS:The mannitol contents in the Cordy ceps samples collected from different growing areas were determined and compared ,and their MDR(Mutual Different Rate)was analyzed.RESULTS:The MDRs among specim ens from Sichuan,Tibet and Yunnan were 0.012~0.32 and the MDRs among the Cordyc eps samples from different micro-ecological environment were 0.037~1.36.CONCLU SION:On the mannitol content in Cordyceps, the effect of the species diversity a nd the micro-ecological environment might exceed that of district and climate d ifference.

ABSTRACT

Object To investigate and compare the scavenging effect of Cordyceps growing in different environment on hydroxyl radical. Methods 10-Phennanthroline-Fe 2+ oxidative assay was used to observe the effect of scavenging hydroxyl radical from H_2O_2/Fe 2+ system by the aquenous extracts of Cordyceps. Results Cordyceps from Naqu of Xizang, northwest Yunnan, and northwest Sichuan all showed us the significant effect on scavenging of hydroxyl radical produced from Fenton Reaction. The biggest values of MDR among Cordyceps specimens from various microecological environment surpassed that among specimens of Naqu of Xizang, northwest Yunnan, and northwest of Sichuan. Conclusion The aquenous extracts of Cordyceps could scavenge the hydroxyl radical from Fenton Reaction. The microecological environment for Cordyceps growing has an significant effect on its scavenging action.
